No. An iTunes card is like a gift card for purchasing media from the iTunes store. iTunes software is free to anybody, whether or not they have an iPod. All you need to purchase media from iTunes is an account (they are free, you just need an email address) and a credit card. If you don't have a credit card and wanted to buy something from iTunes, then you would need a gift card.
And you don't need to buy media from iTunes to put it on your iPod. You can upload music from CDs or link music and videos from other sources on your computer, as long as they are in a compatible format.
