1. George Lucas
2. Steven Speilberg
3. Francis Coppola
4. Walt Disney
5. Martin Scorsese
I would put Walt Disney at #2 on the list, but as Alan pointed out, he hardly directed anything but short cartoons like the Alice Comedies.
George Lucas and Steven Speilberg are two of the best movie directors working today. IMO
However there are alot of great people that should be on that list! Where's : Alfred Hitchcock , Blake Edwards, Brad Bird? All should be on there.
I think Martin Scorsese is hugely overrated. You couldn't say how happy I was when The Aviator lost best screenplay, best director and best picture!

"Greatest" is a little hard to tab. Tim Burton is a director who makes me want to see a movie if he's at the helm. That might make him my favorite. Spielberg has had some misses, but there's no doubt that he's one of the greats. Hitchcock is up there too.
There are directors who aren't necessarily "the greatest" or who I'm not overly familiar with, but have done excellent jobs with certain movies. Alfonso Cuaron is an example- I was very impressed with HP3.

Funny how everybody mentioned Tim Burton because I was originaly going to mention him.
I have one word that really descibes Tim Burton's films: bizzare. I think he has probably the most bizzare imagination in Hollywood. I think Beatle Juice is probably weirdest film ever made. It was just so original and different. I think the movie itself was alright.
I love Nightmare Before Christmas. What surprised me even more is that Tim Burton didn't direct the film. It just seemed so Tim Burton like that it seemed impossible for him not to direct it. It's one of my favorite animated films of all time!
I also think Big Fish is incredibly underrated. I really think that film is wonderful! It was one of the best films of 2003! That's right better than Return of the King. It had such great acting and a script I reall think was well writin. And Tim Burton did a nice job on creating a imaginative world.
Overall Tim Burton isn't one of my all time favorites, but I really think he's made some good films and does a good job directing.

Spielberg's canon is the strongest and more of his films are represented among my favorites than probably any other director. Alfred Hitchcock, David Lean, Francis Ford Coppola, and Martin Scorsese have all made plenty of great films. Among the oft-forgotten comedy directors, I'd have to give praise to the daft John Hughes, Chris Columbus (back when he was directing comedies anyway), and Ivan Reitman. Among young/upcoming directors, M. Night Shyamalan has shown promise, but style often conquers substance and he seems so full of himself that it makes me think less of his films. Wes Anderson's four films are pretty quality. And no one could direct a live action Disney film like Robert Stevenson. Did they ever offer anyone else the job? 
Oh, and John Lasseter and Robert Zemeckis. Plus George Lucas has a handful of good films. Penny Marshall too.

Let's see, who's my favorite director? Hmmmmmmm...
Well, George Lucas is a great director, if not such a great writer. And Tim Burton has a style that I really like. And Steven Speilberg has a great way of getting the audience to really feel for the characters. the problem with lists and discussions like this is that they turn into a 'my dad can beat up your dad' type of thing. If Netty likes David Lynch, so what? He has his right to like him just as Lazario has his right not to.
