Man, this is going to be a hard one for me. Like some have stated already, humor is subjective, and often I enjoy a movie because if makes ME laugh, while others considered to be the funniest ever made just put me to sleep.
Here are some movies that always get a chuckle out of me.
Young Frankenstein, Spaceballs, Robin Hood Men in Tights: Young Frankenstein I first saw back in 2007, and I really loved it. The other two I saw growing up and they never fail to put a smile in my face. I mean, in Robin Hood's case, how can you NOT chuckle at scenes like this one...

Duck Soup: Now this is a movie I recently saw and IT HAD ME IN STITCHES. Seriously, the timing is impeccable and the dialog is mad funny. Even the surreal humor works very well.
Ace Ventura Pet Detective: A bit of a guilty pleasure, this was the first Jim Carrey movie I ever saw and I still laugh at many of its scenes, especially the hospital scene where he dons a tutu.
Airplane!: I really enjoy the visual gags in this one. They always get me with every view.
South Park Bigger Longer and Uncut: There is more to this movie than just shock humor. A lot of it is very clever and with excellent timing. The songs may be some of the funniest ever put in any animated film.

I may not laugh out loud whenever I watch it, but I love
The 40 Year Old Virgin to the point of obsession. I quote this movie with my friends all the time, moreso than Anchorman, Napoleon Dynamite, or any other quotable film of the last 10 years.
I'd post clips or quotes, but I don't want to get in trouble.

Lately, The Hangover is the movie that made me laugh the most.
When I was a kid, I loved Ace Ventura and Dumb and Dumber. I could watched them over and over again. I guess my parents don't like these movies anymore! lol
